Structural Biology Platform Laboratory is mainly divided into the following modules, each module has completely large-scale equipment and mature advanced technology, and each module complements each other to form a complete technical chain of protein structural research. The platform provides the following parts of instrument and equipment support services and technological research and development services:

(1) Protein expression screening module: This module focuses on high-throughput protein expression screening, and quickly realizes the expression optimization of recombinant protein through various host expression systems (such as Escherichia coli., Pichia pastoris, etc.) and expression vectors with various fusion tags. The platform is equipped with high-throughput orifice shaking tables, multi-channel (6/24/96 channels) ultrasonic crushers and other equipment suitable for quickly establishing high-throughput expression condition screening and optimization system. The vectors capable of expressing target proteins can be obtained in 4-6 weeks, the expression condition of target proteins can be optimized and improved in 2-3 weeks, and mass expression of target protein can be delivered in 2-3 weeks.

(2) Protein preparation module: This module uses complete equipment and chromatography packing material library to accomplish large-scale and high-throughput protein expression and preparation. The platform is equipped with high-throughput automatic protein purification systems, preparatory ultracentrifuges, etc., which can realize rapid separation and preparation of proteins and provide milligram to gram preparation services of soluble proteins, inclusion body proteins, membrane proteins and anaerobic proteins.

(3) Protein Characterization Module: This module analyzes the physical and chemical properties of proteins comprehensively, including protein quantification, enzyme kinetics determination, stability analysis and so on. Equipment of this module, such as multifunctional microplate reader, stability analyzer and quantitative identification system for protein expression, can quickly and efficiently characterize the structure and function of protein and improve the research efficiency.

(4) Structural biology sample preparation module: This module is dedicated to the cultivation of high-quality protein crystals and the preparation of frozen electron microscope samples, providing crystallization feasibility evaluation, which can obtain protein crystals of target samples meeting data collection standards within 1-8 weeks. Realizing high-throughput crystal screening and observation by using crystal screening optimization robot and automatic crystal observation system can efficiently obtain samples meeting structural analysis requirements.

(5) Protein structure analysis module: The platform analyzes the three-dimensional structure of proteins by X-ray diffraction, frozen electron microscope and other technical means to reveal their functional mechanism. This module is equipped with a high-performance data processing workstation, which can quickly collect and analyze the diffraction data of protein crystals, and help researchers to understand the interaction between biological macromolecules more deeply. Rapid resolution of protein crystal structures can be realized within one week, and for small molecular structure, it only takes a few hours.
